Thisorangefluorescentphalloidinconjugate(equivalenttoTRITC-labeledphalloidin)selectivelybindstoF-actinswithmuchhigherphotostABIlitythanthefluorescein-phalloidinconjugates.Usedatnanomolarconcentrations,phalloidinderivativesareconvenientprobesforlabeling,identifyingandquantitatingF-actinsinformaldehyde-fixedandpermeabilizedtissuesections,cellculturesorcell-freeexperiments.Phalloidinbindstoactinfilamentsmuchmoretightlythantoactinmonomers,leADIngtoadecreaseintherateconstantforthedissociationofactinsubunitsfromfilamentends,essentiallystabilizingactinfilamentsthroughthepreventionoffilamentdepolymerization.Moreover,phalloidinisfoundtoinhibittheATPhydrolysisactivityofF-actin.Phalloidinfunctionsdifferentlyatvariousconcentrationsincells.Whenintroducedintothecytoplasmatlowconcentrations,phalloidinrecruitsthelesspolymerizedformsofcytoplasmicactinaswellasfilaminintostable"islands"ofaggregatedactinpolymers,yetitdoesnotinterferewithstressfibers,i.e.thickbundlesofMICROFILaments.ThepropertyofphalloidinisausefultoolforinvestigatingthedistributionofF-actinincellsbylabelingphalloidinwithfluorescentanalogsandusingthemtostainactinfilamentsforlightmicroscopy.Fluorescentderivativesofphalloidinhaveturnedouttobeenormouslyusefulinlocalizingactinfilamentsinlivingorfixedcellsaswellasforvisualizingindividualactinfilamentsinvitro.Fluorescentphalloidinderivativeshavebeenusedasanimportanttoolinthestudyofactinnetworksathighresolution.AATBioquestoffersavarietyoffluorescentphalloidinderivativeswithdifferentcolorsformulticolorimagingapplications.

1.Prepare1000XPhalloidinDMSOstocksolution:byadding30uLofDMSOintothepowderformvials.

2.Prepare1XPhalloidinconjugateworkingsolution:byadding1uLof1000XPhalloidinconjugateDMSOsolutionto

1mLofPBSwith1%BSA.

Note1:Theunused1000XDMSOstocksolutionofphalloidinconjugateshouldbealiquotedandstoredat-20oC.protectedfromlight.

Note2:Differentcelltypesmightbestaineddifferently.Theconcentrationofphalloidinconjugateworkingsolutionshouldbepreparedaccordingly.

3.Stainthecells:

3.1   Performformaldehydefixation.Incubatecellswith3.0–4.0%formaldehydeinPBSatroomtemperaturefor10–30minutes.

Note:Avoidanymethanolcontainingfixativessincemethanolcandisruptactinduringthefixationprocess.Thepreferredfixativeismethanol-freeformaldehyde.

3.2   Rinsethefixedcells2–3timesinPBS.

3.3   Optional:Add0.1%TritonX-100inPBSintofixedcells(fromStep2.2)for3to5minutestoincreasepermeability.Rinsethecells2–3timesinPBS.

3.4   Add100uL/well(96-wellplate)ofphalloidinconjugateworkingsolution(fromStep1)intothefixedcells(fromStep2.2or2.3),andstainthecellsatroomtemperaturefor20to90minutes.

3.5   RinsecellsgentlywithPBS2to3timestoremoveexcessphalloidinconjugatebeforeplating,sealingandimagingundermicroscope.
