(Ac-ANW)2R110isaselectivefluorogenicsubstrateforproteasome20S-beta5i.Thenon-fluorescentsubstrategeneratesabrightgreenfluorescentrhodamine110productthathasEx/Em=494/521nm,andcanbeeasilydetectedwithaFITCfilterset.Thisrhodamine110substrateismuchmoresensitivethantheAMC-,AFC-or4-nitroaniline-basedsubstrates.Themostcommonformoftheproteasomeisknownasthe26Sproteasomethatcontainsone20Scoreparticlestructureandtwo19Sregulatorycaps.All20Sparticlesconsistoffourstackedheptamericringstructuresthatarethemselvescomposedoftwodifferenttypesofsubunits;alphasubunitsarestructuralinnature,whereasbetasubunitsarepredominantlycatalytic.Theoutertworingsinthestackconsistofsevenalphasubunitseach,whichserveasdockingdomainsfortheregulatoryparticlesandthealphasubunitsN-terminiformagatethatblocksunregulatedaccessofsubstratestotheinteriorcavity.Theinnertworingseachconsistofsevenbetasubunitsandcontaintheproteaseactivesitesthatperformtheproteolysisreactions.Inmammals,thebeta1,beta2,andbeta5subunitsarecatalytic.Althoughtheyshareacommonmechanism,theyhavethreedistinctsubstratespecificitiesconsideredchymotrypsin-like,trypsin-like,andpeptidyl-glutamylpeptide-hydrolyzing.Alternativebetaformsdenotedbeta1i,beta2i,andbeta5icanbeexpressedinhematopoieticcellsinresponsetoexposuretopro-inflammatorysignalssuchascytokines,inparticular,interferongamma.
