1) Preparea10to20mMROSBrite™stocksolutioninDMSO.Make5to10µMworkingsolutionbydilutingtheDMSOstocksolutionintoHankssolutionwith20mMHepesbuffer(HHBS).
2) Treatcellsasdesired(e.g.,RASMcellsaretreatedwith50-100nMangiotensinIIfor3-5hours).
3) IncubatethecellswithROSBrite™(5-10µM,fromStep#1)for15-30minutesat37ºC.
Note:TheconcentrationofROSBrite™usedvarieswithdifferentcelllines,onewillneedtestwithdifferentconcentrationstogettheoptimaldose.Forcat#16053,onemightuselessconcentrationsuchas0.5-5uM.
4) Replacethedye-loadingsolutionwithHHBSbuffer.
5) Analyzethecellswithaproperfluorescenceinstrument(e.g.,afluorescencemicroscope,flowcytometeroraninvivoimaginginstrument).
