Hypochloriteanion(ClO-)anditsprotonatedform,hypochlorousacid(HClO)arecriticalreactiveoxygenspecies(ROS)inbiologicalsystems.Uncontrolledproductionofhypochlorite(hypochlorousacid)canleadtotissuedamageanddiseasesincludingarthritis,renalfailureandcancers.Inaddition,sodiumhypochlorite(NaClO)hasbeenwidelyusedasableachingagentforsurfacecleaning,odorremovalandwaterdisinfectioninourdailylives.Exposuretolargeamountofsodiumhypochloritecanleadtopoisoningwiththesymptomsofseriousbreathingproblems,stomachirritation,rednessandpainonskinandeye.Therefore,highlyselectiveandsensitivedetectionofhypochlorite(hypochlorousacid)isoftoxicologicalandenvironmentalimportance.Amplite™ColorimetricHypochlorite(HypochlorousAcid)AssayKitoffersasensitiveabsorption-basedassayformeasuringhypochlorite(hypochlorousacid)withhighspecificity.Uponselectivereactionwithhypochlorite(hypochlorous)thecolorlessOxirite™HypochloriteSensorgeneratesastrongcolorproduct.Thecolorsignalcanbemeasuredbyaabsorptionmicroplatereaderat~580nm.WiththisColorimetricHypochlorite(HypochlorousAcid)AssayKit,traceamountofhypochloritecanbedetected.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1.PrepareOxirite™hypochloritesensorstocksolution(20X):

Add500µLofDMSOintothevialofOxirite™hypochloritesensor(ComponentA)tomake20Xstocksolution.

Note:Makesingleusealiquots,andstoreunused20XOxirite™hypochloritesensorstocksolutionat-20ºC,avoidlightandrepeatfreeze-thawcycles.

2.Preparehypochloriteassaymixture:

Add250µLofOxirite™hypochloritesensorstocksolution(fromStep1)into5mLofassaybuffer(ComponentB),andmixwelltomakehypochloriteassaymixture(ComponentA+B).

Note:Thishypochloriteassaymixtureisenoughforone96-wellplate.Itisnotstable,useitpromptly.

3.Prepareserialdilutionsofhypochloritestandards(0to1%):

3.1   Preparehypochloritestandarddilutions:Add100μLofhypochloritestandard(ComponentC)into400μLofassaybuffer(ComponentB)toget1%hypochloritestandardsolution.Perform1:3serialdilutionsinassaybuffer(ComponentB)getapproximately1%,0.3%,0.1%,0.03%,0.01%,0.003%,0.001%and0%seriallydilutedhypochloritestandards.

Note:Theunusedhypochloritestandard(ComponentC)shouldbedividedintosingleusealiquotsandstoredat-20ºC.

 

3.2   Addserialdilutionsofhypochloritestandardandhypochlorite/hypochlorousacidcontainingtestsamplesintoa96-wellclearbottommicroplatemicroplateasdescribedinTables1and2.

4.Runhypochlorite/hypochlorousacidassay:

4.1   Add50μLofhypochloriteassaymixture(fromStep2)toeachwellofhypochloritestandards,blankcontrol,andtestsamples(seeStep3.2)tomakethetotalvolumeof100µL/well.

Note:Fora384-wellplate,add25μLofsampleand25μLofhypochloriteassaymixtureintoeachwell.

4.2   Incubatethereactionatroomtemperaturefor3-5minutes,protectedfromlight.

4.3   MonitortheabsorbanceincreasewithanabsorbanceplatereaderatOD=555±5nm.
