Reactiveoxygenspecies(ROS)arechemicallyreactivemoleculescontainingoxygen.Examplesincludesuperoxide,hydroxylrADIcal,singletoxygenandperoxides.ROSishighlyreactiveduetothepresenceofunpairedvalenceshellelectrons.ROSformsasanaturalbyproductofthenormalmetabolismofoxygenandhaveimportantrolesincellsignalingandhomeostasis.However,duringtimesofenvironmentalstress(e.g.,UVorheatexposure),ROSlevelscanincreasedramatically.Thismayresultinsignificantdamagetocellstructures.Cumulatively,thisisknownasoxidativestress.ROSarealsogeneratedbyexogenoussourcessuchasionizingradiation.Underconditionsofoxidativestress,ROSproductionisdramaticallyincreased,resultinginsubsequentalterationofmembranelipids,proteins,andnucleicacids.Oxidativedamageofthesebiomoleculesisassociatedwithagingaswellaswithavarietyofpathologicalevents,includingatherosclerosis,carcinogenesis,ischemicreperfusioninjury,andneurodegenerativedisorders.ROSBrite™APFisafluorogenicprobetomeasurehydroxylyradicalincellsusingconventionalfluorescencemicroscopy,high-contentimaging,microplatefluorometry,orflowcytometry.Thecell-permeantROSBrite™APFreagentisnonfluorescentandproducesbrightgreenfluorescenceuponreactionwithhydroxylradical.Theresultingfluorescencecanbemeasuredusingfluorescenceimaging,high-contentimaging,microplatefluorometry,orflowcytometry.Inthepresenceofperoxidase,APFalsoreactswithhydrogenperoxide.APFhasgoodselectivitytohydroxylradicalcomparedtootherROS.APFandHPFshowrelativelyhighresistancetolight-inducedoxidation.APFandHPFarenonfluorescentuntiltheyreactwiththehydroxylradicalorperoxynitriteanion.APFwillalsoreactwiththehypochloriteanion.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1)      Preparea10to20mMROSBrite™APF(orHPF)stocksolutioninDMSO.Make1to10µMworkingsolutionbydilutingtheDMSOstocksolutionintoHankssolutionwith20mMHepesbuffer(HHBS).

2)      Treatcellsasdesired(e.g.,RASMcellsaretreatedwith50-100nMangiotensinIIfor3-5hours)

3)      IncubatethecellswithROSBrite™APF(orHPF) (1-10µM,fromStep#1)for20-60minutesat37ºC.

4)      Replacethedye-loadingsolutionwithHHBSbuffer.

5)      AnalyzethecellswithaproperfluorescenceinstrumentatEx/Em=490/525mm(cutoff=515nM)withbottomreadmode(e.g.,FITCfiltersetforafluorescencemicroscope,FL1filterforaflowcytometer).

Note:BSAandphenolredcanaffectthefluorescenceandshouldbeusedwithcaution.BothAPFandHPFcanbeusedinsolutionassaysorforintracellularmeasurements.
