7-AADstainingisnormallyperformedafterallotherstainings.Itstainsdeadcellsonly.Thefollowingprocedurecanbeadaptedformostcelltypes.Growthmedium,celldensity,thepresenceofothercelltypesandotherfactorsmayinfluencestaining.Residualdetergentonglasswaremayalsoaffectrealorapparentstainingofmanyorganisms,causingbrightlystainedmaterialtoappearinsolutionswithorwithoutcellspresent.
1).Make1-10mMDMSOstocksolution.TheDMSOstocksolutionisgoodfor6monthsifstoredat-20°C.
2).Usethefixationprotocolappropriateforyoursample.
3).PelletcellsbycentrifugationandresUSPendthecellsinbufferedsaltsolutionsormedia,withoptimaldyestainingatpH7.4.Adherentcellsinculturemaybestainedinsituoncoverslipsorinthecellculturewells.
4).Add7-AADstainusingtheconcentrationsfrom0.5to5µMandincubateitfor15to60minutesasaguide.
Note:Ininitialexperiments,itmaybebesttotryseveraldyeconcentrationsovertheentiresuggestedrangetodeterminetheconcentrationthatyieldsoptimalstaining.
5).MeasurethefluorescenceintensityatEx/Em=545/650nm(useFL3forflowcytometricanalysis)
CAUTION:7-AADisapotentialcarcinogen.Itisrecommendedthattheuserweargloves,protectiveclothing,andeye/faceprotectioninordertoavoidcontactwithskinandeyes.
