TheHoechststainsareafamilyoffluorescentstainsforlabelingDNAinfluorescencemicroscopy.BecausethesefluorescentstainslabelDNA,theyarealsocommonlyusedtovisualizenucleiandmitochondria.Twoofthesecloselyrelatedbis-benzimidesarecommonlyused:Hoechst33258andHoechst33342.Bothdyesareexcitedbyultravioletlightataround350nm,andbothemitblue/cyanfluorescencelightaroundanemissionmaximumat461nm.TheHoechststainsmaybeusedonliveorfixedcells,andareoftenusedasasubstituteforanothernucleicacidstain,DAPI.ThekeydifferencebetweenthemisthattheadditionalethylgroupofHoechst33342rendersitmorelipophilic,andthusmoreabletocrossintactcellmembranes.Insomeapplications,Hoechst33258issignificantlylesspermeant.ThesedyescanalsobeusedtodetectthecontentsofasampleDNAbyplottingastandardemission-to-contentcurve.

PelletcellsbycentrifugationandresUSPendthecellsinbufferedsaltsolutionsormedia,withoptimaldyebindingatpH7.4.Adherentcellsinculturemaybestainedinsituoncoverslipsorinthecellculturewells.AddHoechststainusingtheconcentrationsbetween0.5and5µMandincubateitfor15to60minutesasaguide.Ininitialexperiments,itmaybebesttotryseveraldyeconcentrationsovertheentiresuggestedrangetodeterminetheconcentrationthatyieldsoptimalstaining.
