Thelong-wavelengthtracernuclearyellowisoftencombinedwiththepopularretrogradetracertruebluefortwo-colorneuronalmapping.Inneuronalcells,nuclearyellowprimarilystainsthenucleuswithyellowfluorescence,whereastrueblueisaUVlight-excitable,divalentcationicdyethatstainsthecytoplasmwithbluefluorescence.BothnuclearyellowandtruebluearestablewhensubjectedtoimmunohistochemicalprocessingandcanbeusedtophotoconvertDABintoaninsoluble,electron-densereactionproduct.BisbenzimideandNuclearYellow,whentransportedretrogradelythroughaxonstotheirparentcellbodies,maymigrateoutoftheaxonsandthecellbodies,asindicatedbyfluorescenceofadjoiningglialnuclei.Thismigrationwasfoundtotakeplacebothinvivoandinvitroduringstorageofthesectionsinwater.Whenusingthetracersin1%concentrationstheinvivomigrationmaybecontrolledbyrestrictingthesurvivaltime.Theinvitromigrationmaybepreventedbyrapidhistologicalprocessingofthematerial.

Thefollowingprocedurecanbeadaptedformostcelltypes.Growthmedium,celldensity,thepresenceofothercelltypesandotherfactorsmayinfluencestaining.Residualdetergentonglasswaremayalsoaffectrealorapparentstainingofmanyorganisms,causingbrightlystainedmaterialtoappearinsolutionswithorwithoutcellspresent.

1)      Pelletcellsbycentrifugation

2)      ResUSPendthecellsinbuffered solutionsormediawithoptimaldyebindingatpH7.4.

3)      Stainadherentcellsinsituoncoverslipsorinthecellculturewells.

4)      AddHoechststainusingtheconcentrationsbetween0.5and5µM.

5)      Incubatethecellsfor15to60minutes.

Note:Theincubationtimeanddyeconcentrationneedexperimentallytested.Ininitialexperiments,itmaybebesttotryseveraldyeconcentrationsovertheentiresuggestedrangetodeterminetheconcentrationthatyieldsoptimalstaining.
