CyberOrange™isanexcellentnucleicacidgelstain,andexhibitslargefluorescenceenhancementuponbindingtonucleicacids.IthasthesamespectralpropertiestothoseofSYBR®Gold,thusagreatreplacementtoSYBR®Gold(SYBR®GoldisthetrademarkofInvitrogen).ItisoneofthemostsensitivestainsavailablefordetectingDNAinagaroseandpolyacrylamidegels.CyberOrangehashighersensitivityforDNAthanRNA,andisidealforusewithlaserscannerswiththesameinstrumentsettingsofSYBRGold.CyberOrangeismuchmoresensitivethanethidiumbromideforDNAinagarosegels,anditcandetectaslowaspicogramdsDNAongels.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1.Post-stainingProtocol

1.1Rungelsbasedonyourstandardprotocol.

1.2Make1XCyberOrange™workingsolutionbydilutingthe10,000XstockreagentintoPH7.5-8buffer (e.g.,TAE,TBEorTEpreferablypH8.0).

Note:Stainingsolutionspreparedinwaterarelessstablethanthosepreparedinbufferandmustbeusedwithin24hourstoensuremaximalstainingsensitivity.Inaddition,stainingsolutionspreparedinbufferswithpHbelowabout7.5orabove8.0arelessstableandshowreducedstainingefficacy.

1.3Placethegelinasuitablepolypropylenecontainer.Gentlyaddasufficientamountofthe1Xstainingsolutionto  submergethegel.

Note:Donotuseaglasscontainer,asitwilladsorbmuchofthedyeinthestainingsolution.

1.4Agitatethegelgentlyatroomtemperaturefor~30minutes,protectingfromthelight.

Note:Thestainingsolutionmaybestoredinthedark(preferablyrefrigerated)foraweekandreusedupto2-3times.
