1.Thawthecomponents:Thawallthekitcomponentsatroomtemperature,andkeepfromlight.
2.Applyanti-fadingreagent:Removeanyexcessliquidfromyourspecimen.Addasmalldropoftheselectedcomponenttothespecimen.Ifthesampleisonaslideortissueculturedish,carefullyplaceacoversliponthedrop,avoidingairbubbles.Ifthesampleisonacoverslip,invertthecoversliponacleanglassslide.Removeanyexcessanti-fadingcomponent.
3.Preparesamplesforimaging:Theanti-fadingreagentsshouldbeincubatedfor2hourstoovernight.Forlong-termstorage,sealthecoversliptotheslidewithnailpolishoraplasticsealant.Mountedslidesshouldbestoredat4ºCinthedarkforoptimumsamplelongevity.Thefluorescenceimagingwouldremainstableformanyweeks.Samplescanbeimagedimmediatelyaftermounting.AtypicalimageisshowninFigure1.
Note:Theseready-to-useanti-fadingreagentscanbeapplieddirectlyonthewashedspecimen.Althoughthereagentshavebeentestedwithlotsoffixedsamples,theiroptimalanti-fadingefficienciesstronglydependonthepropertiesofyoursamples.Wesuggestthatyoutrymorethanonecomponentforyourimagingsamplestogettheidealcomponent.Forexample,onecomponentmaybemorecompatIBLewithafluorescentlabeledantibodyconjugate(oranenzymesubstrateoraspecialmountingspecimensthatcontainlipophilicplasmamembranestainslikeDiI)thananotherone.
