Thefollowingprocedurecanbeadaptedformostcelltypes.Growthmedium,celldensity,thepresenceofothercelltypesandotherfactorsmayinfluencestaining.Residualdetergentonglasswaremayalsoaffectrealorapparentstainingofmanyorganisms,causingbrightlystainedmaterialtoappearinsolutionswithorwithoutcellspresent.
1) PelletcellsbycentrifugationandresUSPendthecellsinbufferedsaltsolutionsormedia,withoptimaldyebindingatpH7.4.Adherentcellsinculturemaybestainedinsituoncoverslipsorinthecellculturewells.
2) MakeaTMA-DPHDMSOstocksolutionat10-50mM.
3) AddTMA-DPHstainusingtheconcentrationsbetween0.5and5µMinHanksand20mMHepesbufferpH7.4,andincubateitat37°C,5%CO2incubatorfor5minutes.Ininitialexperiments,itmaybebesttotryseveraldyeconcentrationsovertheentiresuggestedrangetodeterminetheconcentrationthatyieldsoptimalstaining.
4) WashthecellsandresuspendedintheappropriateHepesbufferpH7.4.
