TMA-DPHisafluorescentprobeformembranefluiditymeasurements.Itisespeciallyusefulinthestudyofmonolayerdynamicsoflipoproteinsandsimilaranalogoussystems.DPHanditsderivatives(suchasTMA-DPH)arecylinder-shapedandhavefluorescenceemissiontransitiondipolesthatarebasicallyalignedparalleltotheirlongmolecularaxis.Assuch,theyareverysensitivetoreorientationresultingfrominteractionswithsurroundinglipids.Theyarewidelyusedinfluorescencepolarizationstudiesofrotationalmotion.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

Thefollowingprocedurecanbeadaptedformostcelltypes.Growthmedium,celldensity,thepresenceofothercelltypesandotherfactorsmayinfluencestaining.Residualdetergentonglasswaremayalsoaffectrealorapparentstainingofmanyorganisms,causingbrightlystainedmaterialtoappearinsolutionswithorwithoutcellspresent.

1) PelletcellsbycentrifugationandresUSPendthecellsinbufferedsaltsolutionsormedia,withoptimaldyebindingatpH7.4.Adherentcellsinculturemaybestainedinsituoncoverslipsorinthecellculturewells.

2) MakeaTMA-DPHDMSOstocksolutionat10-50mM.

3) AddTMA-DPHstainusingtheconcentrationsbetween0.5and5µMinHanksand20mMHepesbufferpH7.4,andincubateitat37°C,5%CO₂incubatorfor5minutes.Ininitialexperiments,itmaybebesttotryseveraldyeconcentrationsovertheentiresuggestedrangetodeterminetheconcentrationthatyieldsoptimalstaining.

4) WashthecellsandresuspendedintheappropriateHepesbufferpH7.4.
