1.PrepareThioliteGreen™workingsolution:
1.1 Preparea10to25mMstocksolutionofThiolite™Greeninhigh-quality,anhydrousDMSO.Thestocksolutionshouldbeusedpromptly;anyremainingsolutionneedbealiquotedandfrozenat‑20oC.
Note:TheunusedThioliteGreenstocksolutionshouldbedividedintosingleusealiquotsandstoredat-20oC,protectedfromlight.
1.2 Preparea2XThiolite™Greenworkingsolution:Onthedayoftheexperiment,eitherdissolveThiolite™GreeninDMSOorthawanaliquotoftheThiolite™Greenstocksolutiontoroomtemperature.Preparea2Xworkingsolutionatthefinalconcentrationrangingfrom100to250µMin20mMHepesbufferorbufferofyourchoice,pH7.ItisrecommendedtouseThiolite™Greenatthefinalconcentrationrangingfrom50to100µMtomeasureThiolconcentrationinsolution.
2.RunGSHAssayinsupernatants:
2.1 Add50µLof2XThiolite™Greenworkingsolution(fromStep1.2)toeachwelloftheGSHstandard,blankcontrol,andtestsamplestomakethetotalGSHassayvolumeof100µL/well.
Note:Fora384-wellplate,add25µLofsampleand25µLofGSHreactionmixtureintoeachwell.
2.2 Incubatethereactionatroomtemperaturefor10to60minutes,protectedfromlight.
2.3 MonitorthefluorescenceincreasewithafluorescenceplatereaderatEx/Em=490/525nm.
2.4 Thefluorescenceinblankwells(withtheassaybufferonly)isusedasacontrol,andissubtractedfromthevaluesforthosewellswiththethiolreaction.
