Fluoro-Jade®C,likeitspredecessorsFluoro-JadeandFluoro-JadeB,willstainalldegeneratingneurons,regardlessofspecificinsultormechanismofcelldeath.Therefore,thepatternsofneuronaldegenerationseenfollowingexposuretoeithertheglutamateagoNIST,kainicacid,ortheinhibitorofmitochondrialrespiration,3-NPA,werethesameforalloftheFluoro-Jadedyes.However,therewasaqualitativedifferenceinthestainingcharacteristicsofthethreeFluorochromes.Specifically,Fluoro-JadeCexhibitedthegreatestsignaltobackgroundratio,aswellasthehighestresolution.Thistranslatestoastainofmaximalcontrastandaffinityfordegeneratingneurons.Thismakesitidealforlocalizingnotonlydegeneratingnervecellbodies,butalsodistaldendrites,axonsandterminals.ThedyeishighlyresistanttofADIngandiscompatIBLewithvirtuallyallhistologicalprocessingandstainingprotocols.TriplelabelingcanbeaccomplishedbystainingdegeneratingneuronswithFluoro-JadeC,cellnucleiwithDAPIandactivatedastrocyteswithGFAPimmunofluorescence.Fluoro-Jade®isaregisteredtrademarkofHisto-Chem,Inc.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1. Tissuesectionsaremountedontopre-treatedslidesandallowedtodryatroomtemperatureovernight,forexample,mountbraintissuesectionsongelatincoatedslidesanddryat50-60^oConslidewarmerfor30minutes.

2. Immerseslidesinbasicalcoholsolution(20mLof1%NaOHto80mLof100%ethanol)for5min.

3. Transfertheslidesinto70%ethanolfor2min,andthenintodistilledwaterfor2min.

4. Incubatein0.06%potassiumpermanganatesolutionfor10min.

5. Rinseslidesindistilledwaterfor2min.

6. Incubatetheslidesin0.0001%solutionofFluoro-Jade®Cdissolvedin0.1%aceticacidfor10minutes.

Note1:Tomake0.0001%solutionofFluoro-Jade®C,firstmakinga0.01%stocksolutionofthedyeindistilledwaterandthenadding1mLofthestocksolutionto99mLof0.1%aceticacidvehicle.

Note2:Theworkingsolutionwasusedwithin2hofpreparation.Thestocksolution,whenrefrigerated,canbekeptforlongperiodsbutshouldbediscardedifthesolutionbecomescloudy.

7. Rinsetheslideswithdistilledwaterfor1minperchangefor3times.

8. Drainexcesswaterontoapapertowel,andair-drytheslidesonaslidewarmerat50^oCforatleast5min.

9. Cleantheairdriedslidesinxyleneforatleast1min(1-5min).

10. CoverslipwithDPX(FlukaorSigma)nonfluorescentmountingmedia.Donotusepolarcoverslippingmedia,suchasthosethatcontainwater,alcoholorglycerol.

Note1:Forbestpreservationofsections,applyaclearcoatofnailpolisharoundtheedgesofthecoverslip,storeinthecold.

Note2:ToprotectFluor-jadefromphoto-bleaching,mixthesolutionsindimlight.Whentheslidesarestaining,wrapthestainingtraysinaluminumfoil.
