1.PrepareNileRedworkingsolution:
1.1 Preparea1mMstocksolutionofNileRedinhigh-quality,anhydrousDMSO.Thestocksolutionshouldbeusedpromptly;anyremainingsolutionshouldbealiquotedandfrozenat<-20oC.
Note:Avoidrepeatedfreeze-thawcycles,andprotectfromlight.
1.2 Preparea200to1000nMworkingsolutionof1XNileRedrightbeforeusebydilutingNileRedDMSOstocksolution(fromStep1.1)inHanksand20mMHepesbuffer(HHBS)orbufferofyourchoice,pH7.Mixthemwellbyvotexing.
2.RunNileRedwithafluorescencemicroscopeoraflowcytometer:
2.1 Treatcellswithtestcompoundsforadesiredperiodoftime.
2.2 Centrifugethecellstoget1-5x105cellspertube.
2.3 ResUSPendcellsin500µLofNileRedworkingsolution(fromStep1.2).
2.4 Incubateatroomtemperatureor37°Cfor5to10min,protectedfromlight.
2.5 ToremovetheNileRedworkingsolutionfromthecells,washthecellswithHHBSorbufferofyourchoice.Resuspendcellsin500µLofprewarmedHHBSormediumtoget1-5x105cellspertube.
2.6 MonitorthefluorescencechangeatEx/Em=552/636nmwithafluorescencemicroscopeoraflowcytometer.
Note1:Foradherentcells,cellscanbewashedwithHHBSorbufferofyourchoice,andloadedwithNileRedworkingsolutiondirectlyinthecellplate.
Note2:Cellscanalsobeprefixed,andthenstainedwithNileRedworkingsolution.
