OurLiveorDead™FixableDeadCellStainingKitsareasetoftoolsforlabelingcellsforfluorescencemicroscopicinvestigationsofcellularfunctions.Theeffectivelabelingofcellsprovidesapowerfulmethodforstudyingcellulareventsinaspatialandtemporalcontext.Thisparticularkitisdesignedtouniformlylabelfixedmammaliancellsinbluefluorescenceforflowcytometryapplicationswithvioletlaserexcitation.Thekitusesaproprietarybluefluorescentdyethatismorefluorescentuponbondingtocellularcomponents.Thefluorescentdyeusedinthekitiswellexcitedwiththevioletlaser(405nmexcitation)tofluorescenceat460nm.Thekitprovidesalltheessentialcomponentswithanoptimizedcell-labelingprotocol.Itisanexcellenttoolforpreservingoffluorescentimagesofparticularcells,andcanalsobeusedforfluorescenceflowcytometryapplications.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1) Add200µLDMSO(ComponentB)intothevialofStainIt™ReactiveDye(ComponentA)tohave500XDMSOstocksolution.

Note:TheunusedStainIt™dyestocksolutionshouldbedividedintosingleusealiquotsandstoredat‑20 ^oC.Avoidrepeatedfreeze/thawcycles.

2) Preparecellsforflowcytometrystainingusing1XHanksand20mMHepesbuffer(HHBS)orsodiumazide-freeandserum/protein-freebufferofyourchoice.

3) WashcellsoncewithHHBSortheazide-andserum/protein-freebufferofyourchoice.

4) ResUSPendcellsat5-10×10⁶/mLinHHBSorintheazide-andserum/protein-freebufferofyourchoice.

5) Add1µLofStainIt™ReactiveDyestocksolution(fromStep1)to0.5mLofcells/assayandmixitwell.

6) Incubatefor20to60minatroomtemperatureor37°C,5%CO₂incubator,protectedfromlight.

Note:Theoptimalstainconcentrationsandincubationtimeshouldbeexperimentallydeterminedfordifferentcelllines.

7) WashcellstwiceandresuspendcellswithHHBSorthebufferofyourchoice.

8) Fixcellsasdesired(optional).

9) Analyzecellswithflowcytometer.
