Probenecidisaninhibitoroforganic-aniontransporterslocatedincellmembranes.Thesetransportersoftenextrudefluorescentindicatorsfromcells,andthereforecontributetopoordyeretention.Thisphenomenonusuallycauseshighbackgroundintheassaysthatrequirethegoodretentionofthedyeindicatorsinsidecells.Theuseofprobenecidtoinhibitthetransporteractivity,andthustoreduceleakageoftheintracellulardyeindicatorsisacommonmethodforreducingfluorescencebackgroundofcalciumassays.Thecommonlyusedfreeacidformofprobenecidrequirestheuseof1MNaOHtodissolveitduetoitspoorwatersolubilityinneutralwater.AATBioquestoffertheconvenientReADIuse™water-solubleandheat-stableprobenecidintheformatofapowder,solutionortablet.Theyareconvenienttouseandareaseffectiveasthefreeacidformatthesameconcentration.Eachvialisdilutedwith90mLbuffertomake2.5mMfinalconcentrationofprobenecidforyourassays.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1 Thaw1bottleofCat.#20062atroomtemperaturebeforeuse.

2 Make2.5mMReadiUse™probenecidbuffer:Add1mlofReadiUse™probenecid25mMstABIlizedaqueoussolutionto9mlofHHBS(1XHank’swith20mMHepesbuffer,pH7.0),andmixthemwell.

Note1:10mLof2.5mMReadiuse™probenecidbufferisenoughfor1plate.AliquotandstoretheunusedReadiuse™probenecid25mMstabilizedaqueoussolutionat<-20^oC,protectedfromlight.Avoidrepeatedfreeze-thawcycles.
