Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1.Preparecellsasdesired.Forexample,preparethegranulocytesat10⁷cells/mLHanksand20mMHepesbuffer(HHBS),add100μLtoapolypropylenetube.

Note: Eachcelllineshouldbeevaluatedonanindividualbasistodeterminetheoptimalcelldensity.

2.Add1-10μLoftheProtonex™Red600-LatexBeadConjugatetothetubeandincubatewithgentleshakingfor30minutesat37˚C.

Note: Eachcelllineshouldbeevaluatedonanindividualbasistodeterminetheoptimalincubationtime.

3.Prepareanidenticalsamplethatisincubatedat4˚Cascontrol.

4.Attheendofthe30minuteincubation,stopthephagocytosisbyadding2mLoficecoldHHBS.Mixandthenwashthecells2timeswithcoldHBSS.

5.ResUSPendthecellsin500μLofcoldHBSS,keepthesamplesat4˚CandanalyzeassoonaspossIBLeatEx/Em570/600nM(TexasRed®filterset).
