1. Prepare~10mMstocksolutionsinDMSO.
2. Loadintocellsintheculturemediumcontaining5-40uMZinquinethylesterorinPBS(withcalciumandmagnesium).
3. IncubatewiththeZinquinethylesterfor15-30minutesat37ºC.Exactloadingconcentration,timeandtemperaturedependonthepurposeoftheexperimentandcelltype,andsowillneedtobeoptimizedexperimentally.
4. WashwithPBStoremoveextracellularremainingdye,andreplacewithPBSormedium.
5. Observeundermicroscopeoranalyzewithflowcytometer.
