Itiswidelyrecognizedthatfluorescentlabelingofcellsisaneffectivemeanstodeterminetotalcellnumbersorhowmanyviablecellsexistinasample.Flowcytometrycombinedwithfluorescentstainingisapowerfultooltoanalyzeheterogeneouscellpopulations.Fluoresceindiacetate(FDA)anditsderivativesarenon-fluorescentmoleculesthatdiffuseintocellsandarehydrolyzedbyintracellularnon-specificesterasestogivefluorescentproducts.Thefluorescentproductscanbeaccumulatedonlyinthosecellsthathaveintactcellmembranes;therefore,deadcellswithleakymembranesarenotstained.TheprecisekineticsofmembranetransportandintracellularhydrolysisofFDAanditsanalogs(suchasCDCFDA)arerelatedtocellularfunctions,thusFDAlabelingcanbeusedformonitoringcellsbyflowcytometryorfluorescencemicroscopy.ThefluorescenceintensityoflabeledcellsbyFDAdyesvariesconsiderablyamongcelllinesandstrains,probablybecauseofdifferencesinintracellularesteraseactivity.CFDAisanamine-reactiveFDAderivativethatcanbeusedtoprepareavarietyofFDAconjugates.
