- Preparecells:
Plate100to100,000cells/wellinatissueculturemicroplatewithblackwallandclearbottom.Addtestcompoundsintothecellsandincubateforadesiredperiodoftime(suchas24,48or96hours)ina37°C,5%CO2incubator.Forblankwells(mediumwithoutthecells),addthesameamountofcompoundbuffer.Thesuggestedtotalvolumeis100µL/well/96-wellplateand25µL/well/384-wellplate.
Note:WetreatedHeLacellswithstaurosporine(SS)for4hoursat37ºCtoinducecellapoptosis.SeeFigure1fordetails. - Preparedye-loadingsolution:
- Preparedye-loadingsolutionbyadding5μLof200XCellbrite™Red(ComponentA)and5μLof200XNuclearBlue™DCS1(ComponentC)to1mLofAssayBuffer(ComponentB).Thedye-loadingsolutionisstableforatleast1houratroomtemperature.
Note:Astheoptimalstainingconditionsmayvarydependingondifferentcelltypes,it’srecommendedtodeterminetheappropriateconcentrationofComponentAandCindividually. - Replacegrowthmediumwith100µL/well(96-wellplate)or25µL/well(384-wellplate)ofdye-loadingsolution(fromStep2.1).
- Incubatethedye-loadingplateatroomtemperatureor37ºCfor30minutesto1hour,protectedfromlight.
- WashcellswithHHBS,PBS,orbufferofyourchoicetwice.Add100µL/well(96-wellplate)or25µL/well(384-wellplate)ofAssayBuffer(ComponentB)intothecells.
- MonitorthefluorescencesignalundermicroscopewithaTexasRedorCy5filterforlivecells,andaDAPIfilterfordeadcells.Thefluorescenceintensitycanalsobeanalyzedusingafluorescencemicroplatereaderwithabottomreadmode.
- Preparedye-loadingsolutionbyadding5μLof200XCellbrite™Red(ComponentA)and5μLof200XNuclearBlue™DCS1(ComponentC)to1mLofAssayBuffer(ComponentB).Thedye-loadingsolutionisstableforatleast1houratroomtemperature.
