1.Preparecells:
Foreachsample,preparecellsin1mLwarmmediumorbufferofyourchoiceatadensityof5×105to1×106cells/mL.
Note:Eachcelllineshouldbeevaluatedonanindividualbasistodeterminetheoptimalcelldensityforapoptosisinduction.
2.Prepare200XThiolite™Green:
Add500µLofDMSO(ComponentC)intothevialofThiolite™Green(ComponentA),andmixwell.
Note:AliquotandstoredtheunusedComponentAat-20oC.Avoidrepeatedfreeze/thawcycles.
3.RunGSHGreen™Assay:
3.1 Treatcellswithtestcompoundsforadesiredperiodoftimetoinduceapoptosis.
3.2 Add5μLof200XThiolite™Green(fromStep2),andincubatethecellsina37°C,5%CO2incubatorfor15to30minutes.
Note1:Foradherentcells,gentlyliftthecellswith0.5mMEDTAtokeepthecellsintact,andwashthecellsoncewithserum-containingmediapriortotheincubationwithThiolite™Greendye-loadingsolution.
Note2:Theappropriateincubationtimedependsontheindividualcelltypeandcellconcentrationused.Optimizetheincubationtimeforeachexperiment.
3.3 Optional:Centrifugethecellsat1000rpmfor4minutes,andthenre-sUSPendcellsin1mLofAssayBuffer(ComponentB)orbufferofyourchoice.
3.4 MonitorthefluorescenceintensitywithaflowcytometerusingtheFL1channel(Ex/Em=490/525nm).Gateonthecellsofinterest,excludingdebris.
