Thefollowingprocedurecanbeadaptedformostperioxidaseandhydrogenperoxidebasedassays.Ininitialexperiments,itmaybebesttotryseveralhydrogenperoxidesolutionsovertheentiresuggestedrangetodeterminetheconcentrationthatyieldsoptimalperioxidase.
1 Prepareaperoxidasereactionmixturewithanappropriatehydrogenperoxidesubstrateconcentrationand50to500µMhydrogenperoxide.
2 Addequalvolumeofperoxidasereactionmixtureandperoxidasestandardsandperoxidase-containingsamples.
3 Incubatethereactionfor15to30minutesatroomtemperature,protectedfromlight.
4 Monitorthefluorescenceorabsorbancewithappropriateinstruments.
HRPdoseresponsewasmeasuredwiththeAmplite™ADHP(Cat#11000),andReadiUse™hydrogenperoxidesolution(cat#11004).
